The analysis of cyanotoxins using LC-MS/MS in Korea is commonly used as the standard method, requiring different standard materials for each type of microcystin, posing an economic disadvantage for total microcystin concentration analysis. By utilizing the common structure of microcystins, the ADDA structure, a method is provided for quantifying total microcystin concentrations. This method involves the conversion of the ADDA structure to 2-methyl-3-methoxy- 4-phenylbutyric acid (MMPB) through oxidation with potassium permanganate and sodium periodate. This study aimed to propose a total microcystin analysis method applicable to water samples, investigating sample concentration and purification methods, as well as the suitability of internal standard materials. Sample concentration was achieved via freeze-drying, while sample purification utilized the liquid-liquid extraction (LLE) method. Internal standards 4PB and MMPB-d3 were evaluated, with 4PB used for instrument calibration and quantification, and MMPB-d3 for oxidation process correction and recovery rate calculation. Although the MMPB analysis method for total microcystin quantification requires careful consideration, its applicability to various matrices and lack of necessity for different standard materials offer advantages. Further evaluation is expected to refine the analysis method. |
