[Introduction]
It has been reported that approximately 270 types of MCs exist in nature, and the current standard method, LC-MS/MS, quantifies each variant using corresponding reference materials. However, the commercially available MC reference materials are limited, restricting the measurement of total Microcystins (Total MCs). MMPB is produced by the oxidation of the Adda (the abbreviation for (all-S, all-E)-3-amino-9-methoxy-2,6,8-trimethyl-10-phenyldeca-4,6-dienoic acid) structure in MCs using potassium permanganate and sodium periodate, enabling the measurement of total Microcystins without the need for individual reference materials.
This study aimed to establish an LC-MS-based quantification method for MCs using MMPB oxidation for water samples. Pre-treatment methods such as pre-concentration, oxidation, clean up and calibrations for liquid samples were compared and evaluated. This study focused on comparing pre-concentration methods, post-oxidation purifications, and the suitability of calibration conditions to develop MMPB quantifications for water sample.
